Production of Glutamic Acid Decarboxylase and Associated Polypeptides for Use in Clinical and Research Applications

Detailed Technology Description
The vast majority of Type 1 diabetes (T1D) patients have sera which contain auto-antibodies and T cells reactive to glutamic acid decarboxylase (GAD) and/or peptide fragments of GAD. The present UCLA invention involves the cloning and production of GAD polypeptides for use in detection of autoantibodies and T cells reactive to GAD in biological samples. This method, therefore, may be developed into antibody and T cell-based diagnostic kits for identifying individuals at risk of developing T1D and to distinguish late onset T1D patients from Type II diabetes patients. Furthermore, the invention may also be used for purposes of screening drugs, such as those that alter GAD function, and for generation of polyclonal and monoclonal antibodies which, in turn, can be used diagnostically to detect GAD.
