A Novel Genetic Method For Generating Gain-Of-Function Mutations
Scientists at UCSD have discovered methods and compositions for generating and using novel overexpression activity alleles of a gene in any organism, especially Drosophilia. Such alleles may be utilized in screening assays, and used to generate dominant-negative forms of bacterial toxins. The method of the present invention provides advantages in efficiency and speed over current methods and is not based on assumptions relating to protein domains or functions, and thus can provide recovery of mutants in unbiased fashion. Although the method was developed for use in Drosophila, it should be applicable to yeast, C. elegans, Arabidopsis and mammals. The method can be used to generate novel mutations in human genes as expressed in test systems to lead to functional determinations and identification of medically relevant partner genes. Dominant-negative forms of bacterial toxins can be designed to interfere with and neutralize natural toxins thereby providing a protective product.
