A novel method for T-cell receptors and immunoglobulin clonotypingusing a new hybrid capture method coupled to DNA sequencing
*Abstract

Through a new hybrid method, the ability toclonotype T-cell receptor (TCR) gene segments and immunoglobulins (Ig) has beenenabled. Current technology relies onhigh-throughput PCR amplification from blood cells carrying target DNA. New clonotyping of TCRs described hereidentify gene targets directly from a library, which also allows for tumorcharacterization in parallel. Furthermore, the new technique is applicable to detection from blood plasma. Thus eliminating the need for extensive PCRamplification and reducing the cost of analysis by 10 – 20 X below currentmethods, leading to expanded capabilities to perform high-throughput analysisat reduced cost. Cell-free applicationsoffer sensitive, less invasive diagnostics with the ability to detect theorigin of DNA from diseased cells from a variety of tissues. Cancer diagnostics and immunotherapy is arapidly expanding sector and would benefit from this novel methodology thatperforms clonotyping and tumor characterization in parallel.
