Determination Of Absolute Configuration Of Secondary Alcohols Using A Competing Enantioselective Conversion Kit

Technology Benefits

(1) Reduces time and labor for the CEC method; (2) Compounds remain stable for many months; and (3) Single-use kits allow for convenient, on-demand testing.

Technology Application

A convenient method for determining the absolute configuration of secondary alcohols.

Detailed Technology Description

Determining the absolute configuration of organic compounds is a challenging problem, but necessary to predict how a compound will react with other chemicals. Several methods for this determination such as x-ray analysis, CD methods, Mosher’s, and VCD with computational analysis are time consuming and labor intensive. The Competing Enantioselective Conversion (CEC) method for determining the absolute stable configuration of alcohols was developed at UCI in 2011. The analysis using CEC is much faster than other state-of-the-art methods, taking only 1-2 hours. Unfortunately, CEC requires a number of solutions with very precise concentrations whose preparation takes much longer than the actual analysis. Furthermore, some of the required chemicals are unstable, preventing the preparation of large batches for future use. For researchers, this means large amounts of samples are collected and the analysis is performed only periodically. Such a drawback is not conducive to time-sensitive development and research. The inventors at UCI have discovered and optimized stable solutions of the reagents necessary for CEC, and prepared a single-use kit for carrying out the analysis. The reagents are prepared in a larger batch that is separated into single-use kits and stored for future use. The kits, which are sealed in an inert gas environment, remain stable for months. Researchers no longer need to wait for a number of samples and can perform the CEC analysis on-demand. Overall, the invention reduces the time and labor for the CEC method and increases its commercial viability. This is particularly important in the Chemical and Pharmaceutical industry.
