*Abstract: This invention discloses novel methods for the enzymatic enrichment and detection of RNA 5' ends that eliminates contaminating RNA sequences and dramatically enriches for RNAs that contain the nuclease resistant 5' cap structure. The invention avoids the tedious and inefficient column purification procedure and can clone mRNA from as little as 500 ng. The cloned sequences are anchored at the 5' end of capped RNAs thus simplifying the bioinformatics analysis to compare samples.
