Real Time Imaging Via Stimulated Raman Spectra Signal

Summary
Researchers at Purdue University have developed a multiplex stimulated Raman scattering (M-SRS) microscope that utilizes a parallel detection scheme to increase imaging speed from one second per pixel to twenty microseconds per pixel. This novel parallel detection device produces high-resolution images with a large signal to noise ratio that can be used to make quantitative measurements with a pixel dwell time of a few microseconds. The system has already been used to produce spectroscopic images of live pancreatic cancer cells that can be processed to parse out the location of lipids, proteins, and water in the cells. All this information will find application in histology, pharmacology, and clinical diagnosis.

*Background
Raman spectroscopy is widely used in biomedical, pharmaceutical, and material sciences due to its high chemical selectivity and noninvasive nature. The limitation of this technique is that the photon scattering being measured is weak, and consequently, Raman microscopy requires a very slow imaging speed. This slow acquisition of data prevents Raman spectroscopy from being used for large area mapping or live imaging of biological systems that are always in motion. A fast imaging method is needed to expand Raman spectroscopy's use in scientific research and clinical practice.
