In This Invention, Two Phytase Genes From Two Generally-Regarded-As-Safe Microorganisms, Bacillus Licheniformis And Bacillus Subtilis 168, Were Cloned And Characterized. A Process For Phytase Enzyme Over-Expression And Purification Was Also Developed. The Enzymes Have Molecular Weight Of About 48 Kilodaltons And Showed Extracellular Phytate-Hydrolyzing Activities. The Recombinant Enzyme Can Be Used To Enhance Phytase Utilization In Various Commercial Areas, Including Preparation Of Animal Feed And Transgenic Plants That Have Increased Growth Rates For Maturity, Flowering And Fruiting.

Summary: (III) is useful for preparing a cell or its progeny capable of expressing a phyL or 168phyA polypeptide which involves transfecting the cell with (III). (IV) or (V) is useful for preparing (I) by recombinant techniques. (VIII) or (IX) is useful for mobilizing inorganic phosphate from plant phytate in a plant cell for improving plant growth, flowering, and/or fruiting, which involves introducing a nucleic acid molecule comprising (VIII) or (IX) into a plant cell to produce a transformed plant cell, whereby the transformed plant cell expresses the phytase which mobilizes the inorganic phosphate from plant phytate. The method further involves producing a whole plant from the transformed plant cell, where the plant comprises the cell that expresses the phytase, and sexually or clonally reproducing the whole plant, where a progeny of the whole plant comprises a cell that expresses the phytase. The expression cassette is introduced into the plant cell by electroporation, microparticle bombardment or microinjection (all claimed). (I) has great commercial utility, since it can be mass produced and incorporated into animal feeds which can be utilized by monogastric animals. This reduces the excretion of unutilized phosphorus into the environment and minimizes environmental pollution. Also transgenic plants incorporating (II) and which express (I) can utilize phosphorus that is otherwise unavailable to plants (phosphorus locked in the form of phytate).
