Summary
The present invention describes an electrochemistry-based real-time polymerase chain reaction (PCR). This invention aims to address the deficiencies inherent in the prior art of optical-based (e.g. fluorescence) real-time PCR method by utilizing the novel strategies based on redox-labeled peptide nucleic acid probe and solid phase extension of oligonucletotide capture probe with redox markers to realize the real time electrochemical PCR technique.

This invention also contains a microchip platform strategy to carry out the electrochemistry-based real-time PCR on a portable chip based system. This electrochemical approach utilizes simple and miniaturizable instrumentation compared to the bulky and sophisticated optics required in the fluorescence-based schemes. This invention can be used for the real-time bioanalyte (e.g. water pathogens, E. Coli) detection in a decentralized environment.
