This technology provides a robust method of synthesizing well-defined and diversely structured high molecular weight polypeptides under mild, open-air conditions, an improvement over the classic method of liquid phase peptide synthesis which requires anhydrous conditions and carefully controlled temperature settings.Ring-opening polymerization of interfacial phase amino acid-derived N-thiocarboxyanhydrosulfide (NTA) monomers into a polypeptide is achieved when the monomers are combined with a polymerization initiator. NTAs are significantly less moisture- and temperature-sensitive than the Ν-carboxyanhydrides (NCAs) monomers used in classic peptide synthesis, making this method suitable for laboratories lacking advanced synthetic facilities.
