Method to Enhance Recombinant Protein Production in Chinese Hamster Ovary (CHO) Cells for Biopharmaceutical Applications

Detailed Technology Description

Chinese hamster ovary (CHO) cells are the mostcommonly used mammalian cell line for production of biopharmaceuticalproteins.  The recent developmentof genome-scale technologies permits direct cell engineering by manipulatingsingle genes that play important roles in metabolic or regulatory pathways togenerate high-producing cell lines. RNA interference (RNAi), induced in mammalian cells via smallinterfering RNA duplexes or short hairpin RNA vectors, negatively regulatesgene expression in a sequence-specific manner.  This technology is comprised of a method to use RNAi toselectively reduce levels of an actin cytoskeletal regulatory protein, cofilin1, to enhance specific productivity of recombinant proteins in CHO cells.
