*Abstract
Nanoparticles (NPs) have been synthesized with the following key characteristics: 1) rod shape, to maximize extravasation to tumor cells via enhanced permeability and retention (EPR) effect; 2) releasable anti-cancer drug to kill tumor cells; and 3) a polymer composition that degrades in the tumor microenvironment (low pH and metalloproteinase activity).Innovations of this research center on the design of a NP drug delivery system that has (i) a unique and effective shape (rod-shape), (ii) encapsulated cytotoxic drugs, and (iii) an ability to release drugs. The elongated rod-shape NP design allows for long circulation time in the body and multivalent interactions with tumor cells (patent pending), increasing the probability of receptor-ligand interactions. Preparation of rod-shape NPs presents a technical challenge. We have overcome this challenge.Another application of PCL NPs is to remove endotoxin from drinking water, biological medium and any medical wares:We have synethsized PCL NPs to remove endotoxin from water significantly. The intellectual merit of this proposal is the design and synthesis of polymeric NPs using an easy, one step and scalable synthesis method (please see methods of PCL NP preparation, p. 4-5), and its efficiency in removing endotoxin.
