Tau-nY29 is a mouse monoclonal antibody (mAb) specific for human tau whereinthe Tyrosine residue at position 29 has been nitrated.
*Abstract

The antibody is of the IgG1 subtype. The antibody detects soluble tau and paired helical filament tau fromseverely affected Alzheimer’s brain but fails to recognize tau from a normalaged brain.

The Tau-nY29 mAb is a powerful tool for detection soluble human tau with anitro group attached at amino acid Tyr29. The nitration of Tyr 29 is indicativeof neurodegenerative diseases, including Alzheimer’s.  Therefore, this antibody is useful instudying the role of nitration of tau in Alzheimer’s disease.

The Tau-nY29 mAb was derived from a hybridoma cell line created through thefusion of SP2/o myeloma cells with splenocytes from a mouse immunized with asynthetic peptide corresponding to human tau amino acids 25-37 that wasnitrated at Tyr29.  The hybridoma cellline was subcloned four times and stored in frozen liquid nitrogen.  Tau-nY29 is specific for human tau constructsnitrated at Tyr29.  It neither reactswith non-nitrated peptides nor human tau proteins nitrated at other tyrosineresidues as demonstrated by ELISA and immunoblot assays.
