*Abstract
BackgroundAfter surgical or accidental trauma, a person first exhibits a strong inflammatory response, but this is then followed by a dramatic depression of cell-mediated immunity. This trauma-induced immunodepression renders the person vulnerable to localized infection as well as generalized sepsis and shock.Nitric oxide synthesis contributes to the early inflammatory response following trauma, however the role of iNOS in the delayed immune dysfunction following severe tissue trauma is unknown. Through the use of in vivo murine trauma models and assessment of immune responses with both genetic and pharmacologic evaluations, investigators have discovered a significant contribution of iNOS in the delayed post injury immunodepression following severe tissue trauma. These findings represent NO as a therapeutic target for immunomodulation to limit injury-induced immune suppression. TechnologyThis invention discloses the use of compounds possessing inhibitory activity on inducible nitric oxide synthase or methods of removing excess nitric oxide such as the use of compounds possessing the capacity for nitric oxide scavenging, in a post-traumatic environment as a method of immunomodulation for a therapeutic benefit.
