*Abstract

"The assay utilizes 96- or 384-well plates lined with nickel-coated beads (His-plate). Recombinant His- Polß-8K is expressed in E. coli and purified to homogeneity. [His-Polß-8K is a recombinant, Nterminal His-tag modified sub-domain of Polß corresponding to the 5’dRP lyase or (8K) domain](5).The protein is bound to the plates via the N-terminal His-tag and unbound protein is washed from the plate. Inhibitors are added, followed by the activated substrate, a biotin-conjugated DNA oligonucleotide that forms a double-stranded hairpin."Available for license as a research tool
