*Abstract

Microarrays have become a widely used tool in biology. Their utility, however, is limited by a failure to achieve uniform deposition of materials on the array and a failure to eliminate any non-specific hybridization to the array. The invention addresses both issues by using a polymer to simultaneously pattern and protect the array surface. Patterning the surface of the array with the polymer promotes uniform deposition of materials, as binding to the array surface can occur only through the openings of the polymer. During hybridization, the polymer protects the surface by preventing non-specific binding to the array surface. Following hybridization, the polymer is removed, which also removes any remaining non-specific signal. By improving uniformity and reducing non-specific hybridization the invention increases the utility and sensitivity of the microarray technique.
