Method for Regulating Apoptosis
- Technology Benefits
- High degree of conservation among fission and fusion proteins Alternative target for therapeutic attenuation of apoptosis Target dynamins and thus prevents cells from dying
- Technology Application
- Treat conditions associated with apoptosis Mycocardial infarction, cancer, Parkinson's disease, amyotrophic lateral sclerosis (ALS), and and neurodegenerative diseases
- Detailed Technology Description
- A method for inhibiting cellular apoptosis through the regulation of mitochondrial fission and fusion has been identified by researchers at the University of California, Davis. The method for identifying small molecule inhibitors of mitochondrial fission fusion is described. Such compounds can be capable of regulating cellular apoptosis via their effects on mitochondrial membrane dynamics. UC Davis investigators have verified compound activities in both yeast and mammalian cell systems. Studies have indicated that one compound acts through the mediation of the mitochondrial fission dynamin related GTPase. In addition, UC Davis researchers have discovered that a small molecule inhibitor of Drp1, the mitochondrial division dynamin, blocks the mitochondrial outer membrane permeabilization in isolated mitochondria, the first step in apoptosis induced by truncated Bid (tBid). The new compound is termed mdivi-1.

Title: Molecules for regulating cell death
Usefulness: Molecules for regulating cell death
Summary: For preparing composition useful for modulating mitochondrial fission in a cell; inhibiting or decreasing apoptosis in a cell and treating condition associated with apoptosis, e.g. Parkinson's disease, amyotrophic lateral sclerosis (ALS), stroke, heart attack, congestive heart failure, transplantation, alcoholic hepatitis, and drug-induced liver toxicity (claimed); decreasing cell or tissue damage; inhibiting mitochondrial fusion used to treat conditions associated with increasing apoptosis, e.g. neoplasia, cancer, and tumor; and also used to identify other molecules that modulate apoptosis by competition assays.
Novelty: New substituted mercaptoquinazolin-4(3H)-one compound useful as mitochondrial dynamin-related guanosine triphosphatase (GTPase) inhibitor for treating condition associated with apoptosis e.g. Parkinson's disease, stroke, heart attack
