Northwestern inventors have created an improvement on structured illumination microscopy that achieves sub-diffractional lateral resolution imaging. This invention builds on existing structured illumination microscopy and tomography technology by combining it with a laser scanning scheme (LSSIM/T). It includes an optical module that can be attached to laser scanning equipment from any manufacturer. The invention encompasses methodology to modulate the laser scanning illumination or excitation temporally, lateral resolution of optical coherence tomography (OCT) that exceeds the diffraction limit, and tuning of the periodic frequency of temporal modulations. This will allow for improved resolution beyond the resolution limit in a variety of microscopy modalities, such as confocal microscopy, non-linear microscopy, optical coherence tomography and others.
