*Abstract

Northwestern researchers have created tissue culture celllines derived from the peripheral blood of a multiple myeloma patient who hadbeen treated with glucocorticoids and become resistant to that treatment.  The blood was separated on a Ficoll gradientto isolate the compartment containing the myeloma cells. These isolated cellswere expanded in tissue culture and named MM.1 cells. The MM.1 parental cellline was further sub-cloned by limiting dilution to isolate cells that aresensitive to the killing effects of glucocorticoids. The resultant cells arenamed MM.1S.  The MM.1 parental cell linewas also treated with glucocorticoids at increasing doses over time and theresultant cells were not killed by high doses of glucocorticoids. These cellsare named MM.1R.  These cell lines havebeen widely used for the pre-clinical development of therapeutics designed totreat multiple myeloma. There are many other human myeloma cell lines inexistence, but these cell lines are unique in that bothglucocorticoid-sensitive and- resistant sub-clones were derived from the samepatient. This is a very attractive model for the study of how glucocorticoidresistance develops in patients.
