Biodegradable Polymeric Buffers

Technology Benefits
The polymers:ΓÇóEnable formation of stable nano-assemblies as compared to conventional systemsΓÇóAllow for a targeted drug/gene delivery with a spontaneous release in the intracellular site of interestΓÇóEffectively protect the encapsulated drug/gene during the deliveryΓÇóAllow for a right charge balance for effective encapsulation, while rendering no or minimal toxicity to non-target organsΓÇóEffectively alter the microenvironment pH of the system, thereby facilitating the endosomal escape of encapsulated drugs/oligo-nucleotidesΓÇóWould be commercially useful for delivery of siRNA/DNA moieties for treatment of cancer and other related disorders

*Abstract
Currently used drug delivery systems (especially for delivery of oligo-nucleotides) involve the use of cyto-toxic complexing agents (cationic lipids and polymers). Additionally, acideic endosomes capture drugs and prevent their release to the proper target. This approach discloses the development and use of novel biodegradable polymers capable of intracellular pH alterations or related buffering actions for an effective drug/oligo-nucleotide release. Such buffers neutralize the pH of the cell and stabilize the drugs to facilitate release from the endosome to the proper part of the cell. The buffer can stand alone as an imaging agent in florescence, or radioactive dyes as well, severing as part of the greater nano systems.
