STRUCTURED VIRAL PEPTIDE COMPOSITIONS AND METHODS OF USE
- Technology Benefits
- HIV/AIDS, viral infections
- Technology Application
- The present invention is directed to compositions, kits and methods utilizing structurally constrained viral peptides. The compositions are useful for treating and/or preventing viral infections as vaccines and to generate antibodies against the structurally constrained peptides that are isolated and used as preventive and/or therapeutic agents.
- Detailed Technology Description
- Hydrocarbon-stapled, alpha-helical viral peptides have excellent structural, proteolytic, acid, and thermal stability, and are highly effective in interfering with virus/cell fusion. A combined strategy of structural data analysis and chemical synthesis was applied to construct membrane proximal external region -based and heptad repeat domain ¬based HIV-1 gp4l immunogens. The invention encompasses the composition and functional validation of gp4l-based stapled peptides, termed Stabilized Antigenic Structures of gp4l (SAS-gp4l), designed to faithfully recapitulate immunogenic features of HIV-1, such that broadly neutralizing anti-HIV-1 antibodies can be elicited upon SAS-gp4l vaccination.

Usefulness: STRUCTURED VIRAL PEPTIDE COMPOSITIONS AND METHODS OF USE | COMPOSITIONS DE PEPTIDES VIRAUX STRUCTURÉS ET PROCÉDÉS D'UTILISATION
Summary: For inhibition or suppression of viral fusion or viral infectivity; for promoting an immune response; for prevention, amelioration, or treatment of a viral infection including HIV infection (claimed) and AIDS; and as vaccine.
Novelty: New structurally constrained peptide comprising specific amino acids of membrane-proximal ectodomain region domain or specific amino acids of heptad repeat domain-2 sequence, for treating viral infection e.g. HIV infection and AIDS
