The production of plasmids containing a nucleic acid sequence encoding a ZmRAD51 fusion protein and their use are described.An isolated protein comprises a polypeptide having at least 90% sequence identity to another polypeptide, wherein the percent sequence identity is determined over the entire reference sequence using the GAP program and they possess recombinase activity. An isolated protein comprises a nuclear localization operably linked to a polypeptide of interest, wherein the polypeptide of interest is localized to the nucleus of a plant cell.The method allows identification and characterization of the functional activities of recombinase enzymes which may offer improved and expanded methods for use in plant systems, particularly agriculturally important crop species such as maize.
