Antibiotic resistance isa growing problem among Gram-positive pathogens. There is thus a need for novel antibacterialcompounds. To date, the two most common strategies for the identification ofnew lead compounds involve either target-based approaches or whole-cellscreening for bactericidal compounds.
Cornell inventors have developedmore specific assays for antibiotics that interfere with cell wall biosynthesiswith two different reporter systems of overlapping specificity. Unlike VanRSsystem that has been used for screening of new antibiotics in drug discovery, theLiaRS system (formerly YvqEC) is not activated by non-specific detergents orother agents that interfere with the cell membrane.
These two-componentsystems LiaRS (for lipid II cycle interfering antibiotic response regulator andsensor) features two regulatory systems from B. subtilis: the sigma factor and the YvqE-YvqC two-componentsystem, both of which coordinate responses triggered by cell wall targetingantibiotics, such as Vancomycin, Bacitracin and Ramoplanin.
