Procedures requiring fixation can be conducted in samples fixed in an identical manner, providing greater uniformity than with paraffin embedded tumor microarrays. Frozen tissue microarray technology allows for analysis of RNA through RNA in situ hybridization, DNA through the use of FISH based experiments and proteins using immunohistochemistry. By allowing simultaneous analysis of uniformly and optimally fixed DNA, RNA and proteins from 100s of tumor samples, this technology may lead to advances in the understanding of tumor pathobiology, and the identification and/or validation of new targets for therapy.

Researchers at the University of California have developed a new method for preparing tissue microarrays using non-fixed, fresh frozen tissue. Sections prepared from these tissue microarrays provide excellent target material for the study of DNA, RNA and proteins as each section can be fixed in a manner specific to the corresponding technique used. Thus, this method circumvents the fixation problems associated with paraffin arrays.

BACKGROUND

Tissue microarray technology is a new method used to analyze several hundred tumor samples on a single slide allowing high throughput analysis of genes and proteins. This technology is useful in that it allows rapid analysis of a large number of samples, as well as simultaneous analysis of different genes and proteins through the use of serial sections. This new technology has already proven useful for rapidly characterizing the prevalence and prognostic significance of differentially expressed genes identified using cDNA array technology as well as genes involved in cancer development and progression. Tissue microarrays have also been useful in identifying genes that are targets of chromosomal amplification as well as to study the expression patterns of putative tumor suppressor genes.

Current methods for making tissue microarrays involve coring tissues from paraffin-embedded tissue donor blocks and placing them into a single paraffin block. One difficulty with paraffin embedded tissue relates to antigenic changes in proteins and mRNA degradation induced by the fixation and embedding process. Consequently, there is a need to identify additional methods that allow for the optimal preservation of DNA, RNA, and proteins in tissue microarrays.
