Chimeric promoters for enhanced gene expression in plants - 1902

*Abstract

 

Overview

Promoters are regions of DNA upstream of the transcriptional start site of a gene where transcription factors bind.  Some transcription factors enhance transcription of the downstream gene while others block its transcription.  Thus, promoters play a key role in the expression of genes. 

 

The exchange of the proximal and distal domains among different native promoter sequences results in recombinant promoters that drive expression of downstream genes.  UK researchers are developing several useful plant promoters using both full-length transcript (Flt-) and sub-genomic transcript (Sgt-) promoters from different members of pararetrovirus. 

 

Invention

UK researchers are developing unique, non-naturally occurring promoters produced from promoters found in Peanut Chlortic Streak Virus (PC1SV), Figwort Mosaic Virus (FMV) and Mirabilis Mosaic Virus (MMV). The activities of these new promoters were determined in both transient and transgenic assays using tobacco plants.  When compared to the CaMV35S promoter, our new sub-genomic transcript promoters (PFlt-UAS-2X, FSgt-PFlt, and MSgt-PFlt) demonstrate demonstrate higher efficacy of successful gene transformation.
