Cornell researchers have identified and characterized two promoters from Coffee arabica: a constitutive alpha-tubulin promoter and an inducible phenylalanine ammonia lyase (PAL) promoter.

Cornell researchers have identified and characterized two promoters from Coffee arabica: a constitutive alpha-tubulin promoter and an inducible phenylalanine ammonia lyase (PAL) promoter.

 

The alpha-tubulin promoter can be used to constitutively express genes in coffee and other crops. Its expression level is comparable to 35S-CaMV.

 

Activation of PAL leads to production of secondary metabolites that are involved in plant defense response, such as lignins, suberins, and a variety of phenolic esters that increase the strength of cell walls. The PAL promoter can be used to express desired trait genes when crops are attacked by pathogens or under other stress conditions.

 

The promoters are important tools for generating coffee or other crops to combat diseases, improve hardiness and increased production.

 

Due to the low genetic diversity of coffee species, the lack of resistance to coffee pest such as the coffee berry borer and the potential of other pathogen infestation, the use of these tools are particularly important for coffee plant transformation.
