TGN38 Rabbit Polyclonal Antibody

*Abstract

Material:
A rabbit polyclonal antibody raised against the C-terminal epitope of canine TGN38 C-terminal epitope. The pAb labels the trans-Golgi network. This antibody was found to have cross species reactivity -- immunofluorescence studies revealed juxtanuclear Golgi staining in human fibroblasts (RF-1), canine (MDCK), lupine (LLC-RK1), and simian (COS) cells.

 

Utility:
This antibody is useful in research concerning protein trafficking through the trans-Golgi network.

 

The trans-Golgi network (TGN) plays a major role in the later stages of the classical protein secretory pathway in higher eukaryotic cells. As the last station of the Golgi complex, the TGN plays a pivotal role in directing proteins to the appropriate cellular destination. Additionally, the TGN plays a role in endocytic pathways that recycle certain receptors between the cell membrane and the TGN.

 

TGN38 is an integral membrane protein that is predominantly localized to the TGN at steady state but can also be observed at the cell surface. Since it cycles constitutively between the TGN and the cell membrane, TGN38 is ideally suited as a model protein for the study of post-Golgi trafficking. Since TGN38 moves through both the endocytic and exocytic pathways during cycling, it has been used as a model protein to identify post-Golgi localization motifs.

 

Deletion of the cytoplasmic tail of TGN38 results in increased expression of the truncated protein at the cell surface, suggesting that this topologic domain contains information necessary for intracellular localization of the protein. Through chimeric protein construct experimentation, it was determined that the cytoplasmic tail of TGN38 has sufficient information for protein localization to the TGN.

 

Dr. Rodriguez-Boulan and his team have developed a rabbit polyclonal antibody raised against a C-terminal of TGN38, CKRSKVTRRPKASDYQRLNLKL, which was frst identified by Wilde A, et al (1992).
