Method to Enhance Recombinant Protein Production in Chinese Hamster Ovary (CHO) Cells for Biopharmaceutical Applications

- Chinese hamster ovary (CHO) cells are the mostcommonly used mammalian cell line for production of biopharmaceuticalproteins. The recent developmentof genome-scale technologies permits direct cell engineering by manipulatingsingle genes that play important roles in metabolic or regulatory pathways togenerate high-producing cell lines. RNA interference (RNAi), induced in mammalian cells via smallinterfering RNA duplexes or short hairpin RNA vectors, negatively regulatesgene expression in a sequence-specific manner. This technology is comprised of a method to use RNAi toselectively reduce levels of an actin cytoskeletal regulatory protein, cofilin1, to enhance specific productivity of recombinant proteins in CHO cells.
