Scientists at UCF have synthesized several fluorescent probes which make use of two photon absorption to monitor pH in biological systems as well as for 3D imaging of cells. These two photon absorbing probes have several advantages: less scattering from cells, deeper penetration of light in samples and the use of near-infrared radiation for excitation. 3D imaging is improved over single photon absorption methods due to reduced scattering and deeper cell penetration. These probes are water soluble and highly effective as an intracellular pH indicator. Intracellular pH is determined by the ratio of fluorescence emission from the probe at two specific wavelengths. The dye has tested to have good permeation into the cell and disperses quickly once inside. Cell toxicity of the probe has been monitored and is extremely low over a wide concentration range.
*Abstract

Monitoring intracellular pH is crucial for understanding the regulation mechanism of many physiological functions, such as: how cells work, biological mechanisms and drug reactions. Understanding these functions is important for curing diseases, improving medicines and the general health and well being of people. Optical methods of determining pH have many advantages over currently available chemical methods, including: rapid response time, being noninvasive, high signal-to-noise ratio and sensitivity. Fluorescent pH indicators that make use of single photon absorption have several limitations: interference or scattering from biological fluids and tissue, photodamage of samples and difficulty in analyzing inter and intracellular pH differences.
