OralImmunization of a live attenuated Escherichia coli strain expressing a holotoxinΓÇÉ structrued adhesinΓÇÉtoxoid fusion protected young pigs against enterotoxigenic E. coli (ETEC)infection

技術優勢
This live vaccine strain expressing an antigen induces antiΓÇÉK88, antiΓÇÉF18, and antiΓÇÉLT immunity. K88 and F18 are adhesins expressing by ETEC strains associated with nearly all postΓÇÉweaning diarrhea (PWD) in pigs. Therefore, this live strain can provide broad protection against PWD. Second, this fusion antigen is constructed in a LTΓÇÉlike holotoxin structure. The antigens can be delivered to the small intestines to induce local mucosal immunity which is believed to play a critical role in protection against enteric diseases.
詳細技術說明
SDSU researchers have genetically fused antigens in a LTΓÇÉliketolotoxin structure to be expressed by a nonpathogenic E. coli isolate for avaccine against PWD. LiveΓÇÉstrain, orally administrated piglets where able todevelop antibodies and protected when challenged with the ETEC strain 3030ΓÇÉ2 while control piglets developed diarrhea and dehydration.
*Abstract
Porcine postΓÇÉweaning diarrhea (PWD) causes major economic losses to swine producers worldwide. Yet, not effective vaccines are available to protect young pigs against PWD. Enterotoxigenic Escherichia coli strains expressing K88ac or F18 fimbriae and heatΓÇÉlabile (LT) and/or heatΓÇÉstable (ST) toxins cause nearly all PWD cases. Vaccines inducing antiΓÇÉK88, antiΓÇÉF18, and antitoxin immunity would provide broad protection against PWD.
