This invention provides isolated riboswitcheslabeled with fluorescent tags, useful for performing single molecule resonanceenergy transfer (smFRET) studies to analyze the dynamics of structural changes in riboswitches. The tagged riboswitches are useful for basicresearch and drug discovery.
*Abstract

Riboswitches are non-coding mRNA elements thatmodulate translation of the mRNA of which they are a part, or transcription ofthe mRNA itself. Structural changes inriboswitches are induced by specific metabolites. Cells have evolved a wide range of use forthis mechanism; changes in riboswitches have activities diverse as haltingtranscription; preventing the mRNA from binding with the ribosome, modulatingalternative splicing, and even affecting translation of other mRNAs. Riboswitcheshave been identified in bacteria, plants and fungi.

 

Cornell inventors have isolated and labeledriboswitches with fluorescent resonance energy transfer (FRET) pairs. Utilizingsingle molecule FRET (smFRET) imaging techniques, the inventors have successfullystudied the conformational changes in several classes of riboswitches, such asS-adenosylmethionine (SAM) riboswitches, thiamine pyrophosphate (TPP)-sensingriboswitches and preQ1 riboswitches. These studies provide insightsinto the mechanisms of riboswitches-mediated gene regulation.
