Benzoxanthene Fluorophores that Emit White Light and Have Favorable Fluorescence Characteristics

This technology is fluorescent xanthene compounds andtheir syntheses. These compounds have applications in biological imaging, andin illumination and display technologies, and they have favorablecharacteristics over the fluorophores already available in market.
*Abstract

A fluorophore absorbs energy at a certain wavelength,and then will release a lower-energy photon, and the difference in wavelengthsof absorption and emission is called the Stokes shift. A large Stokes shiftmakes it easier to use the appropriate instrumentation to separate thefluorescence emission from the scattered illumination. Another desirablequality of a fluorophore is resistance to photobleaching. Fluorescing atmultiple wavelengths help with ratiometric measurements—measuring ratios of theemission peaks.

The synthesis of a benzoxanthene starting from1,6-dihydroxynaphthalene is described. Some of the new compounds have emissionpeaks at three different wavelengths upon excitation at UV, and they emit whitelight. They have many advantages as fluorophores, which are listed below. The synthesisprocedures for the compounds are described in detail. Many spectral propertiesof the compound, SNAFR2, are studied as examples. SNAFR2 is used to labellipophilic components in live HEp-2 cells.

The compound, SNAFR6, has a large Stokes shift (200nm) and fluoresces near IR (~733 nm). (Some of the other compounds also haveemissions in the 500-700 nm range.). Near IR fluorometry of SNAFR6 is performed.The neutral and anionic forms can be excited in the ranges of 420-530 nm and420-650 nm respectively. The anionic emission ranges from 575 nm to 850 nm,with a maximum at 733 nm. It has pKa of 8.18 or 8.10 depending on absorption orfluorescence.
