*Abstract
This invention pertains to a standard operating procedure developed to establish and propagate tissue from the head and neck region of human patients in vitro and in vivo. In order to understand the biology of head and neck mucosa and develop therapeutic agents to treat diseases including cancer of this region, it is imperative to have access to human tissue from normal and diseased mucosa of the head and neck region. The major problem with tissue from the mucosa of the head and neck is that these are typically associated with several micro-organisms including bacteria and fungi. Presence of micro-organisms results in severe contamination of tissue placed in nutrient rich tissue culture media. The contamination limits the availability to good quality tissue for use in vitro and in vivo studies. In order to circumvent this problem, we submerge tissue explants in the operation room in a high dose of antibiotic and antimycotic solution. The tissue is then processed in the laboratory under sterile condition for in vitro culture and implanted surgically in mice for propagation in vivo. The benefit of this method is in the access to fresh human tissue for experimental use. Other applications include access to human tumor tissue that can be tested for drug sensitivity in vitro and in vivo. The drug sensitivity profile could be used to predict which drugs the patient will respond to enabling the physician to make informed choices about the most efficacious drugs for each patient. Thus, this protocol can be used for personalized medicine applications.
