Induction and Isolation of Engraftable Human Spinal Motor Neurons from Human Embryonic Stem Cells

This invention describes a method to selectively induce and isolate spinal motor neurons from human ES cells. Dr. Goldman first induced the HES to differentiate into motor neurons using sonic hedgehog and retinoic acid, then applied the FACS method of D-1997 using a fluorescent protein under the control of the promoter for the moton neuron transcription factor, Hb9. The serial combination of these two steps permits the high-efficiency induction and isolation to purity of functional human motor neurons from cultures of embryonic stem cells.
