Borden, Ph.D.Problem or Unmet Need:Delivering oxygen locally to tissue is important for promoting would healing, treating stroke, enhancing the therapeutic index of radiation therapy and many other applications. Microbubbles are an attractive alternative for fast, non-invasive, intravenous delivery of oxygen. However, current techniques cannot continuously produce oxygen microbubbles at a large scale. This technology presents a method to construct an apparatus and the unique procedure used to continuously generate and accumulate oxygen microbubbles. The apparatus consists of an oxygen source, a lipid source, a pump drive for delivering lipid to the reaction chamber, two large flotation columns, a water cooler to keep the system at a constant temperature, and a sonicator, which produces microbubbles of approximately 2-5 micrometers in diameter. To separate large bubbles from microbubbles, the collection of products from the sonicator are directed into a long flotation column, in which larger bubbles rise to the top and smaller bubbles rise slowly in suspension. The bubbles are removed by syringe and concentrated in a centrifuge to cake form. The bubbles are transferred into glass bottles using a technique requiring three small needles: one for injecting microbubbles into the bottle, another for directing oxygen so the bubbles remain in an oxygenated environment, and the last for venting to maintain pressure equilibrium.
