Method to Alter Chinese Hamster Ovary Cell Line Stability
TECHNOLOGY DESCRIPTION:Researchers at the University of Delawarehave developed a novel tool to increase the genome stability of Chinese HamsterOvary cells, yielding a more reliable and stable process. This new methodtargets the relationship between DSB (double strandedbase) repair system and genome instability in CHO cells,it identifies and expresses related genes to improve the DSB repairand thereby, increasing product stability.APPLICATION: This is apotent tool to improve the genome stability of the CHO cell lines for theproduction of therapeutic proteins. PROBLEMS ADDRESSED: The ChineseHamster Ovary cell lines are most commonly used in the commercial manufacturingof biopharmaceuticals, primarily therapeutic proteins. The protein expressionlevels are low for commercial production, hence the cell limits are pushed byconstant sub-culturing. This renders thecell lines prone to genome instability which further incurs huge losses of timeand money. Thus, there is a necessity for fundamental improvement ofexisting systems. Cell line engineering has great potential for improvingexisting CHO cell expression systems
