Targeted Drug Delivery System for Treatment of Diseases Over-expressing P-glycoproteins
- 详细技术说明
- The invention describes a drug delivery system suitable tooverpass the P-glycoprotein efflux pump. It directly targets the cellsexpressing P-glycoprotein (P-gp); in particular, in certain diseases andconditions such as multidrug resistant (MDR) tumors, neurological diseases, andgastrointestinal disorders.
- *Abstract
-
The innovative featureof this delivery system exploits the expression of P-gp by the cell to tether aliposome, a micelle or a biopolymer as a drug delivery reservoir at the specificsite (tumor site, blood-brain barrier (BBB), gastrointestinal barrier). Thus, the P-gp drug efflux pump is targetedto anchor liposomes to cell surfaces. Then the liposomes gradually release the encapsulated drug.
PotentialApplications
Drugdelivery system targeted P-gp(+) cell types and to be used as:
- Ananti-cancer drug for MDR tumors
- Aneurology or nervous system drug
- Agastrointestinal agent
Advantages
- Targeteddrug delivery system that prevents damage to normal surrounding tissues
- Overcomesthe obstacles of other therapies that aim to suppress or inhibit P-gpexpression.
- Enablespermeation through the BBB and gastrointestinal barrier of drugs loaded in thedelivery system
- Increasesdrug bioavailability
- Reducestoxicity by diminishing the quantity of drug administered
- Very specific binding ability to the targetedsite, and not to normal, healthy body cells, since P-gp also functions as partof the body’s defense against toxic small molecules and therefore should not bebroadly targeted
