Thisinvention describes the synthesis of a chitosan-based precursor that has beenshown to enable various polysaccharide-based hybrid biomaterials, including thepolyethylene amide (PEA), useful in many biomedical applications. The precursors are created by incorporatingglycidyl methacrylate (GMA) into both the hydroxyl and amino pendant groups ofchitosan in the presence of p-toluene sulfonic acid monohydrate,4-(N,N-dimethylamino) puridine and solvent. Unlike previously published methodsof modifying chitosan, the improved synthesis method allows a large degree ofsubstitution, which enables their formulation into hydrogels. In addition, the newbiomaterials are soluble in both water and solvent, and the new method requiressignificantly less GMA and a 50% lower reaction temperature.

 

Awide range of hybrid biomaterials has been synthesized using these new GMA-chitosancore precursors, including integration of polysaccharides with amino-acid PEAsand other biomaterials to create materials with biomedical applications as drugcarriers, growth factors, and substrates for tissue engineering and cellcultures.
