Reagents Useful for Studying Rac/PAK/guanylyl Cyclase Driven Cell Motility

*Abstract

The following reagents are available for licensing:

  • Baculovirus for expression of GST-PAK1 and purified GST-PAK1 protein
  • Baculovirus for expression of GST-PAK2 and purified GST-PAK2 protein
  • Plasmid for expression of His-Rac1 and purified His-Rac1 protein

 

These reagents are useful for screening drug candidates that inhibit Rac-PAK signaling and for studying tumor cell migration and metastasis.

 

Substantial data link the expression and hyperactivity of PAK to tumorigenesis and metastasis. These observations have led to a great interest in understanding the effector pathways downstream of PAK that mediate tumorigenesis and metastasis, and these studies have focused mostly on PAK's classic kinase activity. 

 

Likewise, guanylyl cyclases have been implicated in cell migration. However, how they are activated to promote cell motility has been unknown.

 

Dr. Huang recently described a previously unknown direct communication between guanylyl cyclases and the Rac-PAK signaling pathway through allosteric activation of guanylyl cyclases by autophosphorylated PAK. During this work, they have developed the above reagents for screening drug candidates that inhibit Rac-PAK signaling and for studying tumor cell migration and metastasis.
