Disrupted Adenovirus-based Vaccine Against Drugs of Abuse

- This invention provides an adenovirus-antigen conjugate comprising (a) adisrupted adenovirus with a coat protein or an isolated or purified adenoviruscoat protein and (b) an antigen of the addictive drugs conjugated to theadenovirus coat protein, and a method for using such a conjugate to induce animmune response against the antigen in the human to treat drug abuse.

Addiction to drugs is a majorproblem worldwide, but very little success has been achieved. Vaccinesrepresent a strategy to prevent and treat drug addiction. However, like most small molecules, mostaddictive drugs are poor immunogens and are therefore difficult to developeffective vaccines.
To develop effective vaccines,Cornell researchers have developed a novel vaccine strategy by conjugating an addictivedrug (or an analog or portion thereof) to a disrupted adenovirus with a coatprotein (or to an isolated or purified adenovirus coat protein). Theconjugate is then administered to induce an immune response against theaddictive drug. Adenovirus is an idealcarrier for the antigen of the addictive drugs because adenovirus avidlyinteracts with antigen presenting cells and thus acts as an adjuvant to evokeimmunity against itself.
An anti-cocaine vaccine was designedaccording to the strategy and tested in mice. The anti-cocaine vaccine evoked high titeranti-cocaine antibodies in mice sufficient to completely reverse, on a persistentbasis, the hyperlocomotor activity induced by intravenous administration ofcocaine.

Vaccines against addictive drugs, e.g. opioids and morphinederivatives (cocaine, fentanyl and its analogs, heroin, morphine, opium,oxycodone, hydrocodone), dissociative anesthetics (ketamine, PCP and analogs),depressants (barbiturates, benzodiazepines, flunitrazepam, GHB, methaqualone),cannabinoids (hasish, marijuana), hallucinogens (LSD, mescaline, psilocybin),stimulants (amphetamine, cocaine, MDMA, methamphetamine, methylphenidate,nicotine) and a variety of other drugs including many other prescriptionmedications (e.g. other opioid pain relievers), anabolic steroids, inhalantsand club drugs
