Differentially Expressed Genes Associated with Her-2/Neu Overexpression
The HOMPS genes represent novel targets for the development of diagnostics and therapeutics for breast and ovarian cancer.
Research tools to analyze the role of HOMPS in the pathogenesis of breast and ovarian cancers. May be useful in the development of diagnostics and therapeutics for breast and ovarian cancers.
Researchers at the University of California have identified a new set of proteins, calledHer-2/neu overexpression modulated proteins (HOMPS), based on differential expression in breast cancer cell lines with or without overexpressed HER-2/neu. The HOMPS include proteins having both new and known amino acid sequences. Upregulation of one of the novel proteins, called H41, was confirmed in a human breast cancer specimen in association with HER-2/neu overexpression and in HER-2/neu overexpressing ovarian cancer cells. Polynucleotides that encode H41, polynucleotide probes for the H41 gene, PCR primers for amplifying the H41 gene, and cell lines expressing H41 are available.

Title: Differentially expressed genes associated with HER-2/neu overexpression
Usefulness: Differentially expressed genes associated with HER-2/neu overexpression
Summary: (I), (II) or (X) is useful in diagnostic assays for examining conditions associated with cancer, preferably breast cancer. (I) and (II) are useful for the evaluation and characterization of disorders associated with abnormal cellular proliferation. (I) is useful for generating antibodies for use in detecting HOMPS over expression. (I), its catalytic or immunogenic fragments or oligopeptides are useful for screening libraries of compounds in a variety of drug screening techniques. (X) is useful in imaging methodologies, for purifying HOMPS and mutant HOMPS proteins and polypeptides, and for isolating HOMPS homologs and related molecules. (II) is useful for generating transgenic animals or knock out animals which are useful in the development and screening of therapeutically useful reagents.
Novelty: Isolated human Her-2/neu over expression modulated polypeptide, useful in diagnostic assays for examining conditions associated with cancer

BACKGROUND The human HER-2/neu proto-oncogene encodes a transmembrane receptor tyrosine kinase with extensive sequence homology to the epidermal growth factor receptor. Amplification and/or overexpression of HER-2/neu has been found in one-third of human breast and one-fifth of ovarian cancers. In addition, the HER-2/neu alteration is associated with a poor clinical outcome in that women whose tumors contain it experience earlier disease relapse and shorter overall survival. Considerable circumstantial evidence supports the possibility that overexpression of HER-2/neu plays a direct causal role in the pathogenesis of the malignancies in which it occurs.
