Use of Peptide Hydrogel Scaffolds for Three-Dimensional High Throughput Drug Discovery

- High-throughput screening (HTS) of compound libraries is apromising initial step in building new classes of lead compounds. Cellscultured in 3D matrices presenta more physiologically relevant model and overcome the discrepancies between invitro and in vivo HTSresults. However, most 3D culture technologies have limitations in automation,scalability and reproducibility. The inventionhere describes the novel use of MAX8 b-hairpin hydrogels in 3D matrices for High-throughput screeningthat has been proven to overcome these limitations. The MAX8 peptide and its derivativesundergo assembly at physiological conditions into a hydrogel with awell-defined, nanofibrillar matrix with desired porosity and stiffness and canbe shear-thin injected as a solid material. The self-assembling andhydrogelating b-hairpin peptides possess all thefeatures of an ideal candidate for development as a versatile 3D cell culturematrix that can be dispensed automatically using standard HTS equipment.
