*Abstract

Northwestern University scientists have developed an improved method for prokaryotic cell-free protein synthesis (CFPS) with the ability to incorporate non standard amino acids (NSAA) at defined locations. This cell free platform is derived from cell lysates of an engineered E. coli strain that allows NSAAs to be effectively incorporated into multiple sites and with higher efficiency than the parental strain or other related strains. This system also enables production of proteins requiring disulfide bonds. Additionally, this method enables higher yields, increased solubility, and improved scalability making it faster and more cost effective to produce proteins at an industrially relevant scale. The incorporation of NSAAs provides an endless possibility for the introduction of new functional chemistries at the coding level for a variety of applications in protein and medicinal biochemistry.
