Northwestern researchers designed a novel reporter plasmid that enables research of gene expression in Clostridium acetobutylicum. This bacterial genus is well known for its use in fuel fermentation, but little is known about how solvent production pathways are regulated. To address this problem, Dr. Papoutsakis and colleagues exploited a well-known reporter gene LacZ, whose expression is controlled by three different C. acetobutylicum promoters from the solventogenic metabolic pathways. Expression of LacZ gene is measured by a simple and sensitive β-galactosidase assay, which is routinely performed in many laboratories around the world. In addition, the strength or activation of any other clostridial promoter can be tested in the same manner.
