ES cell system to identify Dnmt1 cytosine methyltransferase mutants that epigenetically reprogram the mammalian genome

*Abstract: DNA methyltransferase 1 (DNMT1) is the most common DNA methyltransferase in mammals and isresponsible for adding methyl groups to DNA which in turn alters gene expression. Investigators at the University of Pittsburgh have developed a system to identify mutant DNMT1 proteins utilizing a genetically engineered mouse embryonic stem cell line. This system has already been shown to identify mutant proteins in that are defective in maintaining genomic imprintingApplications:1) This system may identify mutant proteins that could reprogram a differentiated cell into a stem cell2) This system may identify mutant proteins that could reprogram a cancer cell into a normal cell
