Detection of Hepatits B virus mutations
Worldwide, it is estimated that 400 million people are chronically infected with hepatitis B virus (HBV). Chronic hepatitis B (CHB) infection is the commonest cause of liver cirrhosis and hepatocellular carcinoma (HCC), with an estimated 500,000‐900,000 death per year. Continuing HBV replication increases the risk of progression to cirrhosis and HCC. Thus, antiviral therapy aiming at blocking virus replication is essential in managing CHB patients. A number of nucleos(t)ide analogues such as lamivudine, adefovir dipivoxil, telbivudine, entecavir and tenofovir are approved for antiviral treatment.
Unfortunately, HBV variations frequently develop with long‐term antiviral therapy. Some of these variations confer drug resistance, resulting in treatment failure which may further lead to hepatitis reactivation and hepatic decompensation. Periodic HBV viral load quantification is commonly used to monitor antiviral treatment response and early detection of drug resistance. This invention discloses a method for direct detection of important HBV variations.
